Manchuela D.O. lies in eastern Castilla-La Mancha, spanning around seventy municipalities across the provinces of Cuenca and Albacete. The vineyards sit on a plateau between the Júcar and Cabriel rivers, at altitudes ranging from 600 to 1,100 metres, notably higher than the neighbouring La Mancha plains. Soils are clay over a limestone base, which retains rainfall moisture. The climate is continental, with hot, dry summers, cold winters, low rainfall and the influence of easterly Levante winds, producing a wide gap between day and night temperatures that favours slow, even ripening. The native bobal grape is the appellation's emblematic red variety, alongside macabeo for whites. Other authorised red varieties include tempranillo (cencibel), cabernet sauvignon, garnacha, monastrell, merlot and syrah, while authorised white varieties include chardonnay, sauvignon blanc, verdejo, albillo and viognier.

Manchuela D.O. produces mainly red and rosé wines from bobal, the native grape that gives the appellation its character, alongside white wines based on macabeo. The reds show a deep colour, concentrated, fleshy fruit and firm tannins, with aromas of dark berries such as blueberry and blackberry and spicy notes of liquorice; the best examples can age well. The rosés, bright strawberry in colour, offer aromas of red fruit and violet with a lively, easy-drinking palate. The whites, made from macabeo and other authorised varieties such as chardonnay, sauvignon blanc or verdejo, are light, aromatic and fresh. The reds suit grilled red meats, game and mature cheeses; the rosés pair well with charcuterie, tapas and Mediterranean dishes; the whites are enjoyable as an aperitif or with fish and seafood.
