The appellation lies in Puglia, in the heart of the Salento peninsula, covering the communes of Salice Salentino, Veglie and Guagnano in the province of Lecce, and San Pancrazio Salentino and Sandonaci in the province of Brindisi, together with parts of the communes of Campi Salentina and Cellino San Marco. A limestone bedrock of Cretaceous origin underlies deep clay-limestone soils rich in organic matter in the inland areas where the vineyards are concentrated. The north-south orientation of the rows and the natural ventilation of the Salento plateau favour a balanced growing cycle under a warm, sunny Mediterranean climate. The leading grape is Negroamaro, grown in the area since antiquity and predominant in the vineyards; it has traditionally been accompanied by Malvasia Nera. White wines are based mainly on chardonnay, complemented by pinot bianco and fiano, while aleatico gives the appellation its own sweet and fortified sweet wines.

Salice Salentino produces red, rosé and white wines, together with sparkling versions of the whites and rosés and a sweet or fortified sweet aleatico. The reds show a ruby robe of varying intensity, turning garnet with age; the nose, marked by Negroamaro, recalls ripe red fruit; the palate is full, dry or off-dry, robust yet velvety and warm. Aged at least twenty-four months, six of them in wood, they may carry the Riserva designation and then develop spicy notes. The rosés, with a more or less deep pink colour, offer a delicate fruity nose, sometimes of rose and cherry, leading to a fresh, harmonious palate. The whites, straw yellow with greenish hints, are delicate and fruity when young. The reds pair with red meats and grilled dishes, the rosés suit Mediterranean fare and fresh cheeses, the whites accompany fish and seafood, while aleatico is served at the end of a meal with dry pastries or chocolate.
